South Africa and Australia meet in Durban on Friday for the first in a five-game series that will decide the best one-day side in the world.
Australia relinquished top spot to the Proteas after losing 4-1 at home earlier this year.
Skipper Graeme Smith, who averages 42.21 in ODIs against Australia, is back for South Africa.
Batsman Jacques Kallis misses out with a groin injury, while Philip Hughes is absent from Australia's squad.
The 20-year-old opening batsman passed 50 three times in his first six Test innings against South Africa, finishing the recent series with an average of 69.
Kallis, who hit a century in the third Test against Australia last month, is expected to be back for Sunday's second match, while Morne Morkel and Makhaya Ntini have also been added to the squad.
Twenty20 squad members Yusuf Abdulla, Robin Peterson, Justin Ontong and Johann Louw drop out.
For Australia, Shane Harwood and David Warner have returned home, while Marcus North and Ben Laughlin will be aiming to make their ODI debuts during the series.

The hosts lost the recent Test series 2-1, but hit back in the Twenty20 series, prevailing 2-0.
South Africa also beat Australia 4-1 in the one-day series in Australia in January, and coach Mickey Arthur is hopeful his side can repeat the trick on home soil.
"There's a nice air around the squad," Arthur said. "The guys are very fresh and looking forward to it. We're getting a nice balance now.
"Within our squad, we have two recognised spinners and we have JP Duminy, who can bowl for us as well, so we're starting to get quite a nice mix in terms of variation in our attack.

"And the return of Makhaya [Ntini] and Morne [Morkel] in our attack is really good because they give us variation of a different kind.
"It's always good to be in a dressing room that's winning - Friday is where we want to put our marker in the sand, and that's why we've been building up hard this week.
"Friday, for us, is huge. We want to go 1-0 up, and hopefully take momentum from that."
Smith said he was looking forward to locking horns with Mitchell Johnson again, despite the Australia paceman breaking bones in both Smith's hands in recent months.
"Mitchell has been bowling really well," said Smith. "He is one of the best fast bowlers in the world at the moment and it will be a real challenge facing him. It is something I look forward to.
"We have had a really good battle recently, I have had a bit of success and he has had a bit of success, so we will see what happens."
Johnson said: "It's always good to get the opportunity to bowl with the new ball against an opening batsman like Smith.
"He's such a great player. I'm looking forward to trying to get his wicket."
