Jerome Taylor announced his arrival in England in spectacular fashion with three wickets in 12 balls as the Lions crumbled in reply to West Indies' 203.
The paceman, who only arrived this week after injuring his hip in a car crash, trapped Rob Key leg before first ball.
He dumped Ian Bell on his backside with a bouncer and took two wickets in three balls to leave the Lions 13-3, but Tim Ambrose's 49 helped them reach 143-6.
Earlier, Chris Woakes added two wickets to finish with a career-best 6-43.
The 20-year-old Warwickshire seamer, who took 4-26 on the first day as the Windies stumbled to 76-4, had Shivnarine Chanderpaul caught behind and Brendan Nash pouched by Samit Patel in quick succession to leave the tourists reeling at 91-6.
But captain Key kept his pre-match promise to "give everyone a fair crack" by removing Woakes after a six-over spell.
West Indies added a further 112 runs with David Barnard stroking a useful 58 before falling leg before to spinner Adil Rashid to end their innings shortly after lunch.
The Lions reply got off to the worst possible start when Key played all around a quick swinging delivery from Taylor to depart for a golden duck.

New batsman Bell was greeted with a vicious bouncer that he did well to avoid.
Taylor, who claimed five wickets to help bowl England out for 51 during the first Test at Sabina Park in February, then removed Stephen Moore and Jonathan Trott.
Opener Moore fended off a short ball to Devon Smith at second slip before Jonathan Trott drove at a wide delivery and was caught behind by Denesh Ramdin.
Bell and Patel stabilised the innings but their 44-run partnership was cut short when the former was adjudged lbw to seamer Barnard when on 27 with the ball seemingly missing leg stump.
Patel then top-edged a pull-shot and was caught by Lendl Simmons for the same score to leave the Lions on 78-5.
But Ambrose, who hit eight boundaries in 16 balls either side of tea, guided the home side to within 60 runs of the tourists when bad light brought a premature end to the day's play.
