Rose, 29, carded a six-under 64 and is seven under for the tournament, one shot clear of Australia's Jason Day and South Korean Charlie Wi.
Tiger Woods just made the halfway cut after finishing with an even par 70 to record a three-over 143 halfway total.
"I've just got to put together two good rounds and see where that leaves me," Woods said afterwards.
"Greg (Norman) always said if you started six back on Sunday you could still win and that's certainly within reason."

Woods shot a respectable even-par 70 in the second round on the demanding Aronimink course but missed a putt from barely two feet at the eighth.
"If you don't make putts, no matter how good you hit the ball, you're not going to shoot good scores," he added.
"For some reason today I couldn't release the blade and couldn't get the toe moving. I have to work on that."
Woods' fellow American Jeff Overton picked up three birdies but dropped a shot on his final hole for a second successive two-under 68 to move to four under for the tournament alongside Charley Hoffman in a tie for fourth.
