Durham's bowlers made good use of the pink ball under the floodlights to bowl the MCC out for just 162 on day two of the season curtain-raiser in Abu Dhabi.
Steve Harmison took two wickets in his opening spell as the MCC slumped to 6-3 and never recovered.
Scott Borthwick claimed 4-27 and Callum Thorp 3-25 as the MCC crumbled.
Durham, who had earlier declared on 459-9, with Kyle Coetzer eventually out for a career-best 172, did not enforce the follow-on and closed on 7-2.
Aberdeen-born opener Coetzer resumed on unbeaten on 123, with his side on 329-3.
He lost overnight partner Ian Blackwell early on but then enjoyed an 81-run fifth-wicket partnership with teenager Ben Stokes, who made 51 on his first-class debut for the county.
Coetzer passed his previous best of 153 not out against Durham UCCE in 2007 but his resistance was finally ended when he became one of four late victims to the part-time leg-spin of Dawid Malan after a marathon eight-and-a-half hour, 371-ball stay at the crease.
But the MCC's hopes of challenging the county champions' total were dealt a major early blow as Harmison removed both David Sales and highly-rated Leicestershire youngster James Taylor in a fiery opening spell after Thorp had trapped Scott Newman leg before.
MCC captain Alex Gidman tried to steady the ship with Malan, but fell to a leg-side catch by Phil Mustard to give Stokes his maiden first-class wicket.
Malan and James Foster then added 61 for the fifth wicket but both fell to Thorp in quick succession, before Borthwick ripped through the tail, removing Tim Murtagh and Jon Lewis with successive balls.
Durham opted to bat again and sent Borthwick out to open with captain Will Smith.
But the experiment failed to pay off as Durham slumped to 0-2 when Steve Kirby had Borthwick caught behind to complete a pair, before trapping Dale Benkenstein leg before with his very next ball.
Durham batsman Kyle Coetzer told BBC Radio Newcastle:
"It's without a doubt the longest knock of my life, but Michael di Venuto has done that plenty of times.
"I've seen him spend the entire game on the field and that's something I would like to try and do a bit more often.
"It's great to get my highest score but when we get home conditions are going to be more bowler friendly so I'll have to get stuck in when we get back."
